Nonparametric vs Parametric Tests of Location in Biomedical Research

Course Overview:
This course addresses the debate on the applicability of parametric statistics to non-normal data distributions.
Empirical research suggests the Mann-Whitney test generally has greater power than the t-test except when data are normally distributed.
Objectives:
Compare the relative power of Mann-Whitney and t-test or ANOVA/ANCOVA.
Examine the distribution of your primary endpoint.

Findings: The change between baseline and post-treatment data has often a normal distribution.
Generally, T-test or ANOVA/ANCOVA outperformed Mann-Whitney, particularly when log-transformed data were included.
Methods: Simulation studies were conducted using polynomials to model five non-normal distributions for baseline and post-treatment scores. The power of Mann-Whitney and T-test or ANOVA/ANCOVA was compared across various conditions, including sample size, correlation, and type of treatment effect.
Conclusion: ANCOVA is preferred for analyzing randomized trials with baseline and post-treatment measures, though in extreme cases, it may be less powerful than Mann-Whitney. In such instances, the treatment effect estimates from ANCOVA should be interpreted with caution.
